Purpose

The Protein Science Facility (PSF) provides users with expertise in a wide variety of protein production and protein engineering services. It provides equipments and techniques to teams who wish to develop experimental strategies for functional analyses of proteins. The facility is certified ISO 9001 :2015.

PSF has recently joined forces with the research group ANABIO-MS (ANAlysis of BIOmolecules by Mass Spectrometry) located at ISA (Institut des Sciences Analytiques). The two sites have complementary expertise in mass spectrometry. This collaboration will promote technological developments on mass spectrometry in Lyon and extended our service offering with cutting edge technology.

Services

The Protein Science Facility (PSF) is organized in three units for:

 

  • Production, purification and quality control of recombinant proteins (quantities > mg) and peptide synthesis.
  • Proteomic analysis by mass spectrometry and molecular interaction studies by surface plasmon resonance (SPR).
  • Structural protein characterization by circular dichroism and 3D structure determination by x-ray crystallography.

 

Production and purification of recombinant proteins :

The facility provides pure and homogenous protein preparations from recombinant plasmids or cloned genes, as well as peptide synthesis.

Equipment: 3 automated systems of purification (AKTAs Purifier, PURE, NGC), Multitron incubators and other biophysical equipment, as well as a CEM Liberty One System for peptide synthesis.

 

Protein characterization by mass spectrometry and protein-protein interaction studies:

The protein characterization unit provides mass spectrometry and molecular interactions studies (by surface plasmon resonance (SPR) for instance).

With 3 mass spectrometers, the unit can develop analytical protocols for the identification, characterization of post-translational modifications, differential quantification of proteins and molecular mass measurement.

The unit accepts samples from a wide range of preparations and concentrations.

For SPR studies, a Biacore T200 is available with or without assistance. Customized services are also available. The Biacore T200 is a polyvalent system, which allows extensive intermolecular interaction studies by surface plasmon resonance of unlabeled molecules, tagged or not. The fields of application vary widely from studies of molecular mechanisms and the relations between structure and function, to the discovery of new biotherapeutical molecules, their development and quality control.

 

•  Structural protein characterization by determination of secondary and three-dimensional structure (circular dichroism and x-ray crystallography, respectively):

X-ray crystallography can determine 3-D protein structures at high resolution. The PSF Structural Biology unit offers crystallogenesis services on automated equipment with or without assistance. PSF can run programs of structural biology from recombinant plasmid to crystal diffraction tests. Data processing and structure analysis are performed in collaboration with a crystallographer associated with the project. PSF also offers protein analysis by circular dichroism.
