« The strategy allowing E. coli to survive within macrophages:  mixing single cell phenotypic heterogeneity and collective behavior »

Adherent Invasive Escherichia coli (AIEC) strains recovered from Crohn's disease lesions survive and multiply within macrophage’s phagolysosomes, an environment that prevents commensal E. coli multiplication. We use single-cell analysis, genetic dissection, genomic assays and mathematical models to monitor the growth status and cell cycle regulation of intracellular AIEC. We observed that AIEC, may replicate or undergo non-growing phenotypic switches. These switches result from successive stringent and SOS response inductions. Although non-growing bacteria benefit from protection to stresses, they are not sufficient to maintain AIEC’s niche on the long-term. To form intracellular bacterial communities, where replication is possible, AIEC need to express pathogeny islands and subsequently build a biofilm like matrix. We observed the persistence of these biofilmed communities for several days and their transfer from  dying cells to new macrophages. Therefore, we propose that intracellular communities provide a reservoir for bacteria and contribute to chronic inflammation.
