1-Immunophenotyping and cell-based assays

 

PRESENTATION

The AniRA-ImmOs facility provides a set of services enabling the study of the immune system through various in-vivo , ex-vivo and in-vitro tests . The platform offers the possibility of carrying out various conventional or novel, controlled and validated tests in order to characterise the immune system (phenotype and function).

AniRA-ImmOs offers expertise and technical support for in-depth analysis of the tests performed. The unique feature of the AniRA ImmOs platform is its ability to combine several type of tests on the same sample, thus considerably increasing the power of biological analysis. This multiparametric approach is possible thanks to close collaboration with other AniRA platforms as well as with local research teams specialised in different fields. The AniRA-ImmOs platform also offers to carry out, in collaboration with researchers, the development of new immunological tests as well as technology transfer.

AniRA-ImmOs is integrated into the regional AniRA platform labeled IBiSA, member of the national Research Infrastructure Celphedia.

 

ACTIVITIES

The AniRA-ImmOs platform offers various services including: 

- A multiparametric and quantitative analysis of immunological populations, in blood or tissues (i.e. spleen, lymph nodes, thymus, bone marrow, peritoneal lavage, MALT, etc.). We have developed, validated and implemented screening tools for an in-depth analysis of the mouse immune system

- A wide range of immunological cellular tests:

  • Cell proliferation test
  • Viability test
  • In vitro cell stimulation
  • Custom immunophenotyping (flow cytometry)
  • ELISA, ELISPOT, FluoroSpot assay
  • Cytokine assay using LUMINEX technology, etc.

 

- Preclinical animal pathophysiological models allowing the study of immune responses:

  • Infectious models (e.g.: Influenza, Vaccinia virus, L. monocytogenes, etc.) allowing the evaluation of the capacity to develop an antibody response, the capacity to develop a specific response of CD8+ T lymphocytes, the capacity to survive a sublethal infection (challenge test)
  • Inflammatory models (e.g., DSS-induced colitis or contact hypersensitivity, etc.) that allow testing of the impact of gene mutation on the inflammatory response in mice.

 

AniRA-ImmOs also assists researchers and offers services to characterise the immune system of genetically modified mice at steady state or during challenge.

 

EQUIPMENTS

The AniRA-ImmOs platform has several pieces of equipment available for independent use upon training:

  • Luminex Bio-plex MAGPIX
  • Elispot/Fluorospot Reader (IRIS Mabtech)
  • BSL-2 equipped laboratory
  • etc...

The AniRA-ImmOs platform uses for its activity all the equipment available on the AniRA-Cytometry platform (conventional, spectral, mass, image flow cytometers) and the equipment of the AniRA-PBES platform (animal physiopathological models)
