2-Real-time metabolism assays
PRESENTATION
Agilent Seahorse analyzers measure the oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of living cells in real-time in 8-, 24-, or 96-well plates. OCR and ECAR are key indicators of mitochondrial respiration and glycolysis, as well as ATP production rates. The four-port injection system with automatic mixing allows for real-time analysis, in living cells, of the cellular response to different substrates, inhibitors, and other compounds. Standard metabolic tests are offered, such as ATP production rate measurement, mitochondrial stress testing, glycolysis stress testing, etc. The temperature-controlled system allows studies on a wide variety of cell samples.
The Agilent Seahorse XF HS Mini analyzer measures OCR and ECAR rates in real time for limited quantities of living cells in 8-well plates. A new design concentrates the cells to be analysed in the centre of the wells, thus providing greater measurement sensitivity with very few cells. This equipment also allows measurements to be carried out in confined spaces.
The 24-well plate format allows for the analysis of larger and/or metabolically highly active samples. Metabolic function measurements can thus be performed on cultured cells, ex vivo samples (tissues, islets, biopsies, etc.) and small organisms (nematodes, zebrafish embryos, fruit flies, etc.).
The 96-well plate format allows for the analysis of multiple experimental conditions at once, allowing for flexible assay design, dose-response studies, and screening. Metabolic function measurements can be performed on cell lines or primary cells, adherent or in suspension, as well as on isolated mitochondria.
Biotek's Cytation 1 multi-mode microplate reader combines brightfield cell imaging with quantitative fluorescence detection. The multi-mode detection module features high-sensitivity filter fluorescence and a monochromator for UV-Vis absorbance. The device is temperature-controlled. An integrated solution with the Seahorse 24 and 96 analysers is offered, allowing the combination of cell analysis and quantitative imaging intuitively to normalise data and streamline data interpretation.
ACTIVITIES
The service is open to the academic and industrial scientific community.
Different levels of service are offered: independent or assisted use of equipment, or full service provided upon quotation and after a feasibility study. All consumables are provided. The service also provides advice and assistance for project implementation, adapting technology to a specific context, data analysis, and interpretation.
Regular independent use of the instruments requires free but mandatory training. Any new will be required to agree to the internal policies to gain access to the online booking system.
