3- Protein production and peptide synthesis

 

PRESENTATION

The biochemistry platform allows the production of pure and homogeneous proteins, for functional or structural studies. Recombinant protein expression is performed in a bacterial system only. Purification steps are carried out by chromatography, and various quality controls are performed to finalise the production. Experiments can be performed by the facility staff or by the user themselves.

The centre also offers a peptide synthesis service and an automated immunoassay service on the JESS system.

 

SERVICES

Different levels of services are offered: full services or independent use of equipment upon training

1. Full support for a protein production project

From the recombinant plasmid, expression and solubility control, expression in bacterial system, purification by chromatography or other, dosages and quality control by TSA, NanoDSF or on biophysical equipment of circular dichroism, SEC-MALS and DLS 

 

2. Peptide Synthesis Service

Synthesis of up to 70 residues with a wide choice of scales from 0.05 to 5 mmol, with different post-translational modifications (biotinylation, chromophore, 13C and or 15N labeling, cyclisation, etc.). In the case of small proteins, peptide synthesis can be an alternative for the production of around ten mg, at a lower cost.

 

3. Immunoassay Service on JESS

A sensitive, reproducible and quantitative immunoassay in 3 hours! This is a capillary migration system (cartridges of 25 capillaries) for separation according to 3 ranges (2-40 kDa, 12-230 kDa, or 66-440 kDa) with chemiluminescence or fluorescence detection. Possibility of quantification, multiplexing in the same capillary. The sensitivity is much better than in classic western blot (3 µg of proteins). More than 3000 primary antibodies are validated by the system, allowing them to be used in optimal conditions.

 

4. Provision of biochemistry equipment 

Mandatory training for the use of equipment, provision of consumables. Agreement is required before independent use.

  • 1 BioSprint 15 robot (Qiagen): mini purification of proteins on magnetic beads for expression, solubility, and purification tests (15 tests possible in parallel)
  • 1 Fusion Fx CCD camera (Vilber Lourmat): allows you to photograph gels, blots and other development media
  • Nanodrop for protein and nucleic acid assays
  • 2 Multitron Incubators (Infors)

  • 1  Typhoon  fluo-phosphoimager  (Cytiva): multi-purpose laser scanner, which allows the precise detection and quantification of samples of proteins, DNA, RNA, or molecules labelled by fluorescence or radioactivity.
  • Liberty One  (CEM) with 2 liquid chromatography chains  HP1100  (Agilent) and  Ultimate  (ThermoScientific)
  • JESS (ProteinSimple  Biotechne)
