AniRA – AGC

CERTIFICATION

AniRA-Genetic and Cellular Analysis is integrated into the regional AniRA platform labeled IBiSA, member of the national Research Infrastructure Celphedia. The Genotyping department of the AGC platform is   ISO 9001:2015 certified

PRESENTATION

The AniRA-AGC (Genetic and Cellular Analysis) platform is divided into 3 departments:

                    - the AniRA Genetic Analysis service,

                    - the AniRA Genotyping service,

                    - the AniRA CRB CelluloNet service

1- The AniRA Genetic Analysis aim is to provide support in Quantitative PCR techniques. Real-time PCR is a powerful and essential tool for the quantification of nucleic acids. It serves a wide variety of applications including gene expression, miRNA expression but also SNP genotyping (allelic discrimination) or copy number variation (CNV). qPCR is also used for the detection and quantification of microorganisms or the characterisation of known or unknown mutations by high-resolution melting (HRM).

2- The AniRA Genotyping service ( ISO 9001:2015 certified )

The Genotyping department, in conjunction with the Mouse Experimental Biology Platform (AniRA-PBES), provides PCR genotyping of transgenic mice. Widely used by members of UMS 3444-US8, this department handles a large number of analyses, representing more than 20,000 PCRs per year.

3- The AniRA CelluloNet service is a cell line banking service that offers the preservation and provision of cell lines. The collection currently includes more than 550 cell lines.

The scientific advisory board of the AGC platform:

The mission of the scientific advisory board is, in close consultation with the facility manager, to provide expert advice on new technology, to assess user needs, to monitor staff workload and career development, to assess equipment usage. The scientific advisory board of the AGC platform is composed of:

- Gérard Benoit, LBMC

- Denis Gerlier, CIRI

- Yann-Gael Gangloff INMG

- Suzy Markossian IGFL

- Antoine Marçais CIRI

-Lionel Ballut IBCP for direction UAR3444

ACTIVITIES

1- The AniRA-Genetic Analysis platform:

The platform manages and provides quality-controlled equipment.

The instruments are complementary and allow:

  • Quantification and control of nucleic acids (Nanodrop, Dropsens, Bioanalyzer).
  • Robotic preparation of 96-well QPCR plates (Tecan Qiagen robots) and QPCR circuits.
  • Drawing of primers and analysis of results using suitable software.

Training/expert support  :

  • The platform offers several training courses to learn real-time PCR quantification technology.
  • The platform provides assistance in setting up projects, interpreting results or adapting a technology to a particular need.

Technology development monitoring and implementation :

  • QPCR is a constantly evolving technology: the platform ensures monitoring of new developments and regularly proposes seminars presenting the most significant innovations.

2- Services of the Genotyping service:

  • Carrying out PCR genotyping for monitoring genetically modified lines.
  • Technology transfer: adaptation of a protocol for genotyping a new line.

3- Services of the CRB CelluOnet service:

  • Receiving new cell lines  :

The CRB CelluloNet aims to collect, as a priority, all new cell lines developed by the contributing teams of SFR BioSciences in order to ensure their traceability and conservation.

  • Availability of cell lines, listed in a Catalog (currently being updated, available soon)

The lines are available to academic laboratories or non-profit research laboratories, subject to compliance with the deposit contract to which each line belongs.

CelluloNet provides a series of quality controls for distributed cell lines to meet the growing demands of scientific journals:

  • verification of adequate cell growth upon thawing,
  • absence of contamination by mycoplasma,
  • cell line authentication: https://www.igna.fr/
